Abstract:
In sexual assault samples, biological mixtures comprised of epithelial and sperm cells are the source of DNA. To obtain individualized profiles, mainly for the sperm cells, this mixture must be separated. A differential extraction procedure is employed to preferentially lyse the epithelial cells (female fraction), followed by the sperm cells (male fraction) to release the DNA of interest. As this process is time consuming, the purpose of this research was to evaluate a simple modification that would reduce the digestion time for the sperm fraction in the differential extraction procedure. Four sets of mixed samples of different semen dilutions and different digestion times were evaluated against a standard 120 minute incubation period. For the preliminary study, which eliminated the female fraction, the 1:1 and 1:100 dilution sets at 1 and 10 minutes, respectively, had significantly less DNA than that of the 120 minute time period within their respective dilution set. For the full differential extraction, all samples at all digestion times had similar yields of DNA compared to that of the standard 120 minute period. The findings demonstrate that it is possible for the lysis period of sperm cells to be shortened without compromising DNA yields.
