Abstract:
With the rise of the nation's opioid epidemic, there is critical need for elucidating a well understood biological mechanism of the neuronal circuitry involved in addiction. We have generated a tool that allows for real time optical detection of opioid neuropeptide release using cell based neurotransmitter fluorescent engineered reporter (CNiFER) technology expressing the delta opioid receptor. CNiFERs are human embryonic stem cells stably expressing a certain GPCR coupled to the Gq proteins, and a FRET based calcium sensor TW2B. Through in vitro characterization, the DOR CNiFER shows subnanomolar specificity for Leu-enkephalin, and a temporal resolution of seconds. This DOR CNiFER will be able to provide insight on local opioid neuropeptide activity in vivo.
