Experimental autoimmune uveoretinitis (EAU) is an animal model of the human condition of uveitis. Several retinal antigens have been implicated in the immunopathogenesis of EAU; the two main antigens are S antigen, a soluble protein and P antigent a particulate protein. Retinal S antigen, a soluble retinal protein that induces experimental autoimmune uveoretinitis (EAU), was purified from bovine retinae. The purification procedure consisted of ultracentrifugation, precipitation by ammonium sulphate, ion exchange chromatography and chromatofocusing; the method was simple and efficient. Retinal S antigen isolated in the absence of protease inhibitors in the purification buffer migrated as a doublet on sodium dodecyl sulphate polyacrylamide gel electrophoresis, with protein bands of 55,000 and 53,000; when protease inhibitors were included in the purification buffer, the retinal S antigen appeared as a single band of molecular weight 55,000. The isolated antigen had an isoelectric point of 5.5, and on Immunoelectrophoresis produced a single precipitin arc against anti-bovine S antigen serum and rabbit anti- bovine retinal extract serum. On double immunodiffusion a single precipitin arc with total identity to bovine retinal extract and partial identity with guinea pig retinal extract, was obtained with purified bovine retinal S antigen against rabbit anti-bovine S antigen serum. The purified antigen stained as a glycoprotein, and no staining was observed with a lipid stain. Chemical cross-linking studies suggested that the protein exists in vivo as a monomer. Experimental autoimmune uveoretinitis was induced in guinea pigs and rats using varying doses of purified retinal S antigen and complete Freund's adjuvant H37Ra; the disease was highly reproducible. The pathogenicity of the purified retinal S antigen was confirmed as the animals developed a predominantly posterior uveitis, with little or no anterior segment involvement. Two strains of guinea pig were tested for their susceptibility to S antigen induced uveoretinitis, Duncan-Hartley and ABX/Peru
