The work in this thesis is concerned with the feasibility of vaccinating foals against one of the most common and pathogenic of the equine helminths, Strongylus vulgaris, by using radiation-attenuated third stage S. vulgaris larvae L3). In order to examine the effect of irradiation on the ability of larvae to migrate and establish in the anterior mesenteric artery an irradiation dose titration experiment was carricd out. Freshly harvested third stage S. vulgaris larvae were irradiated at 90 Kr and 110 Kr in a gamma 60Co irradiation unit before being administered to foals reared under worm-free conditions. The foals showed no clinical or haematological changes and at post-mortem examination the mesenteric arterial lesions were mild and the numbers of fourth stage larvae recovered were low. The ability of larval preparations irradiated at 90 Kr and 110 Kr administered to worm-free foals to induce an immune response capable of protecting these foals was investigated in a vaccination experiment. Worm-free foals were double vaccinated with doses of 250 and 500 irradiated L3 given 3 weeks apart and challenged 4 weeks later with 750 normal s. vulgaris larvae. Vaccinated foals showed no clinical or haematological changes attributable to either the vaccine or the challenge infection whereas worm-free challenge control foals given 750 normal larvae showed typical pyrexia, anorexia and colic 7-10 days after challenge. At post-mortem examination the vaccinated foals had mild to moderate arterial lesions from which reduced numbers of fourth stage larvae were recovered whereas the challenge control foals had severe arterial lesions of arteritis and thrombosis from which up to 25% of the challenge dose of larvae could be recovered.
