The thesis presents an account of past and present classifications of the anaerobic corynebacteria, a survey of the literature on their growth, morphology and cultural characteristics together with their biological properties as vaccines. It was found that certain characteristics of the anaerobic corynebacteria were subject to variability despite close control on their growth, testing and vaccine preparation. A capsular material was found to be associated with early logarithmic phase organisms. This material was composed mainly of lipid and polysaccharide and becomes detached from the bacterium towards the end of its growth cycle. Chemotactic activity of the bacteria resided mainly in this capsular material and was associated with the lipid content which is the first description of a lipid chemotactic factor. This lipid also caused a rapid short-lived activation of phagocytes as measured by carbon clearance at day 2 which was the early peak of the biphasic response caused by whole bacteria producing their second peak at day 14. The polysaccharide fraction of the capsular layer was responsible for hypertrophy of liver, spleen and lungs which in mice peaked at day 14 and was also mitogenic for human T lymphocytes. The cytoplasmic fraction from anaerobic coryneforms had virtually no biological activity whereas cell walls were active in most respects including colony stimulating activity. All five serological groups of anaerobic corynebacteria were biologically active in affecting the lymphoreticular system within the parameters tested, the strain P. avidum II 4982 being particularly active. The study also established that anaerobic coryneform bacteria can cause cell fusion in a direct manner. Supporting research established the basis of lipid induced chemotaxis and activation of the reticulo-endothelial system, the occurrence of lipid chemotactic factors in other bacteria and the first record of lymphocyte chemotaxis and its relevance to anaerobic coryneforms.
