Myasthenia gravis is a disease in which a group or groups of skeletal muscles become quickly fatigued by exercise. Whilst any muscles may be involved, those of head and neck, especially the external ocular muscles, are most frequently affected. The disease may occur at any age and in either sex but typically is a disease of young adult females. It is believed that the muscle weakness is probably due to a pre-synaptic lesion at the neuromuscular junction and that the disease has an autoimmune basis. Serious immunological study of myasthenia gravis began in the middle 1950s and since then intensive investigations have been performed in many centres on various aspects of the immunological abnormalities present in this disease. The work described in this thesis falls naturally into the following groupings; serological (Chapters 3 and 4), attempted induction of myasthenia gravis in experimental animals (Chapter 5), quantitative histological assessment of myasthenic and control thymus glands (Chapters 6 and 7) and finally fine structural investigation of the myasthenic and non myasthenic thymus gland (Chapter 8).
