Investigations have been carried out into the effects of cytochalasin (CB) on tissue culture cells and virus-cell interactions. In PtK-1 cells, CB induces gross cytoplasmic retraction, creating bunches of microvilli on the 11 surface, aerial bridges between cells, and a "frilly edge" at the cellbsubstrate boundary. Scanning electron microscopy permits the study of cells as a population and transmission electron microscopy affords more detailed information about the unusual morphological features induced by the drug. A hypothesis is put forward for the method of cellular retraction, which accounts for the observed abnormal features. Although it has been suggested that CB exerts a primary effect on microfilament systems, ultrastructural studies reveal a lack of effect in PtK-1 cells but the disappearance of microfilament arrays in drug-treated Vero cells. This differential response of cell types is emphasised by the t of CB on macromolecular syntheses. In PtE-1 cells, BIT 21 C13 cells and L cells, DNA synthesis is inhibited to varying degrees, RNA synthesis is inhibited to 40% of the normal level and protein synthesis is affected only in C13 cells. The inhibition of RNA synthesis in FtK-1 cells is due, in part, to a reduction in precursor uptake. From the observations made on morphology cell metabolism in the presence of CB, and in agreement with other workers, a membrane site of action of CB is proposed. The hypothesis that the inhibition of pino- and phago-cytosis by CB will discriminate between viruses using different modes of entry to the cell, is not supported by the evidence. The yields of Vesicular Stomatitis virus (VSV) and adenovirus are unaffected, while those of Vaccinia virus and EEC virus are slightly stimulated in the presence of CB. Although the plaguing of VSV, Vaccinia and Semliki Forest virus (SFV) are unaffected by CB, the number of poliovirus plaques is stimulated in a dose-dependent fashion. The optimum conditions under which CB promotes the mass enucleation of HeLa and Vero cell populations are determined - drug concentration, temperature, speed and time of centrifugation, etc. A variety of cell types are investigated for their potential as producers of enucleate cells - each cell line has its own characteristics of enucleation, which are not readily manipulated to improve the resultant enucleate population. Drug-treated Vero cells consistently yield better than 99% enucleates which, apart from the lack of nuclear material, have a normal ultrastructure. Enucleate Vero cells are capable of respreading in CB-free medium and of maintaining protein synthesis for several hours post-enucleation. Their lifetime is prolonged by incubation at 31°C, rather than 37°C. V3V, SFV and poliovirus all replicate successfully in enucleate Vero cells.
