Friend mouse erythroleukaemia cells are induced by growth in the prescence of 1.5% DMSO to undergo erythroid differentiation. In order to attempt a genetic analysis of Friend cell erythropoiesis I have selected cells for reistance to MSO. Cells were treated with the mutagen MS and cloned in the prescence of DMSO. The majority of cells terminally differentiate in the prescence of DMSO, however a few cells grow and form macroscopic clones. The results of a Fluctuation Test were consistent with a mutational origin for clones resistant to MSO. However EMS did not increase the frequency of clones in DMSO. About half the clones picked from DMSO proved unstable and became inducible by DMSO after culture in the absence of IMSO. Some possible technical reasons for this instability have been investigated, but it is possible that a mechanism for temporary resistance to MSO does exist. All the DMSO resistant clones showed karyotype changes but there was no clear correlation between karyotype and stability or instability. The stable IMSO resistant clones were recloned twice in DMSO before further study, None of the stable "third round" MSO resistant clones accumulated globin mRNA in response to MSO. However, when tested with other Friend cell inducers, some clones were not inducible by any of the agents tested, while others responded to one or more inducers, but not to MSO. Both terminal differentiation and the induction of haemoglobin synthesis are resistant to MSO in all the clones tested: however it has since been shown that spectrin accumulation is inducible by 33VISO in one of the clones, but not in the others. Thus a variety of clones of different phenotype can be obtained by cloning Friend cells in the prescence of MSO. Their properties may provide information on the mechanism of Friend cell inducers, and the coordinate control of erythroid function. Two lines have been fused with an inducible Friend cell parent. All the hybrid clones were found to be inducible by DMSO. If this is due to the genetic recessiveness of the DMSO resistant variants, the variants might be analysed further by complementation testing. Another non-inducible variant, Fw, which was not obtained by a deliberate programme of selection, has also been studied. This line is not inducible by DMSO, but was found to synthesize globins in response to the combination of haemin and DMSO. Thus it is conceivable that the non-indueibility of this cell line is secondary to a defect in haem synthesis. Biochemical investigations utilising globin cDNA found no evidence for the control of globin gene expression by haemin at the level of translation, or of nuclear-cytoplasmic transport.
