Expression and characterization of human butyrylcholinesterase mutants:.
Organophosphorus compounds (OPs) are highly neurotoxic anticholinesterase agents. Their widespread availability and potency make them attractive for use in chemical terrorism and warfare. Developing safe and efficient methods for counteracting the effects of OP intoxication remains a priority. When given prophylactically, the mutant of butyrylcholinesterase (BChEG117H ) is able to hydrolyze OPs before they reach target organs. However, its extensive use as a treatment for OP intoxication is limited by several factors, including its extremely low substrate-turnover rate. To address this problem, we hypothesized that changes in structure of BChEG117H via surface loop modifications (BChEG117H-ENX) would increase the enzyme's catalytic efficiency. Data on butyrylthiocholine substrate hydrolysis is presented for BChEWT, BChEG117H, and BChE G117H-ENX to analyze the efficacy.
Role of cytokines in the immune system of Anopheles gambiae:.
Stress responsive peptides (SRPs) are recognized as insect cytokines that take part in physiological processes such as innate immunity and growth retardation. The peptides induce hemocyte spreading in vitro, an initial hemocyte response against infection in many insect species. A. gambiae is a major vector for Plasmodium parasites that cause malaria. Understanding the mosquito immune system may provide new means to disrupt the natural vector-parasite association. Insect hemolymph contains critical components of the humoral and cellular defense. In this work, we collected hemolymph and whole body mRNA samples from naive, PBS-, and live E. coli-injected larvae and adults and confirmed infection. The RNA samples were analyzed by qRT-PCR for antimicrobial peptide (AMP) transcript levels. The hemolymph samples were separated by SDS-PAGE and subjected to LC-MS/MS analysis. These experiments are parts of the projects that assess the differences in hemolymph proteomes of control and immune challenged adults and larvae.
