The insect genomes encode two Perilipin (PLIN) proteins, PLIN1 and PLIN2. The proteins have been the target of few studies in insects. However, genetic studies in the fruit fly Drosophila melanogaster and biochemical studies in Manduca sexta have shown that PLIN proteins affect the rates of lipid accumulation and degradation. In order to study the roles of PLIN proteins in the metabolism of lipids and fertility of Aedes aegypti mosquitoes, we first cloned cDNAs encoding the protein sequences of PLIN1 and PLIN2. The cDNAs were used to express and purify the proteins and to produce rabbit polyclonal anti Aedes PLIN1 and PLIN2 antibodies, which are an essential tool to investigate the expression of the proteins under different developmental and/or nutritional states. The anti PLIN1 antibody detected a protein which was not predicted by the available genomic information. The complete PLIN1 cDNA sequence was obtained and a new PLIN1 gene model was produced. The fat body expresses both perilipins, whereas PLIN2 is mostly expressed in ovaries. The expression of PLIN1 and PLIN2 in fat body increases when starved adults are sugar fed and the increase correlates with the accumulation of triglycerides. Blood fed mosquitoes show clearly distinct patterns of expression for PLIN1 and PLIN2. PLIN1 is highly expressed in the middle of the vitellogenic process triggered by the blood meal. Most of the lipids are transferred to the ovaries during this time frame. On the other hand, PLIN2 is expressed at the beginning and ends of the vitellogenic process. The expression of PLIN2 in ovaries shows a marked increase a few hours before the uptake and accumulation of TG takes place and remains high until oviposition.
