The Gram-positive anaerobic cocci have long been a large group of loosely classified organisms. Traditionally all clinical specimens have been called Peptostreptococcus and often classification goes no further. The aims of this project were to apply a variety of methods to the study of Gram-positive anaerobic cocci including fast atom bombardment-mass spectrometry, gas-liquid chromatography, thin layer chromatography and partial 16S rDNA sequencing to gather information about the many species in this category which has been previously uninvestigated with a view to improving strain identification. Data regarding the phospholipids, fatty acids, fermentation acids and genetic sequences were collected by growing and harvesting anaerobically grown bacterial cultures. Data were collected from 15 species from seven genera including Peptostreptococcus, Finegoldia, Micromonas, Schleiferella, Ruminococcus, Slackia and Staphylococcus. Data were also collected from five species of Streptococcus and two species of Gemella for comparative purposes. For fast atom bombardment-mass spectrometry, thin layer chromatography and capillary column gas liquid chromatography the phospholipid content of freeze dried bacterial cells was extracted using chloroform and methanol prior to further chemical derivatisation. For the study of fermentation acids using packed column gas liquid chromatography cultures were grown in GLC broth in which the fermentation acids were retained. For partial 16S rDNA sequencing cultures were harvested from broth culture and the DNA extracted using Qiagen DNA extraction kits before being subjected to PCR. Results show primarily that this is such a diverse group of organisms that no two methods gave the same groupings of species. Huge amounts of data were composed regarding the biochemical and molecular composition of each species most of which were data never before collected for these organisms. In conclusion the methods used generated much interesting information about these organisms but it was not possible using these methods on these organisms to create any meaningful taxonomic groupings.
