The Africanized honeybee (Apis mellifera) is one of the most remarkable invasions in the Americas that also resulted in a single event of introduction in Puerto Rico, an oceanic island in the Caribbean. The Africanized bees on the island established successfully and replaced the very much reduced population of European honeybee (Apis mellifera ligustica) on the island. Several studies of the population of bees in Puerto Rico conclude that A. mellifera is a single hybrid population on the island, with an African maternal lineage, and an introgression of European alleles of 35 %. The population is resistant to the Varroa mite and demonstrates a docile behavior, characteristics that distinguish it from continental bee populations. The unique state of Apis mellifera in Puerto Rico is described in the coined term gAHB (gentle Africanized Honey Bee). This population may serve as the model to examine in detail the structure of an invasive population, the origin of the invasive population, and the mechanisms and processes that have emerged during more than 15 years of his incursion, such as change in the defense genes involved in the docile behavior. The gAHB in Puerto Rico presents a defensive behavior so different from the American continent that makes it a good candidate for beekeeping, especially now that the Apis mellifera populations in the world are facing yearly losses. This project focuses in three objectives: 1. Determine the genetic structure of the population of gAHB in PR and examine if gAHB is a genetic mosaic. 2. Determine the source of the gAHB in PR. 3. Compare and correlate gene expression level across aggressive colonies vs. non-aggressive colonies identified within gAHB populations, and examine whether these defensiveness-related genes change expression levels in response to manipulation or presentation of nocive stimuli. My goal is to understand how the genetic characteristics of gAHB are related to the behavioral characteristics of honey bees in Puerto Rico. Answering this question has two implications; it would reveal mechanisms for fast evolution as well as the gene combination responsible for their gentle and resistant behavior. The population analysis of gAHB confirms that its structure corresponded to a single hybrid population, that represents in parts of its genome a mosaic of mostly European or mostly African alleles. I demonstrated that the AHB that invaded Puerto Rico in 1994, came from Texas. Lastly, these results suggest that the expression of aggressive behavior in honeybees is regulated in conjunction with neuromodulators (DAR2, OAR, 5HT-R, TRH) and changes in the metabolic mechanisms of oxidative phosphorylation (ND51 and ND20-LIKE). The results from this project may lead to designing better strategies for the conservation and management of the gAHB, critical for the recovery of bee populations in general. Also, this study contributes to the understanding of how processes of colonization and dispersal of invasive populations can cause drastic changes in population characteristics.
