Desmosomes are intercellular junctions found in epithelial cells, cardiac myocytes, meninges and follicular dendritic cells. They are composed of several proteins, including two glycoproteins, the desmocollins and desmogleins. The glycoproteins are subfamily members of the cadherins, which are a family of calcium dependant cell adhesion molecules known to play a role in cell-cell adhesion and embryogenesis. The desmosomal glycoproteins exist as three distinct isoforms, which are the products of single genes (Dsc1, Dsc2 and Dsc3, Dsg1, Dsg2 and DsgS). These isoforms are differentially expressed in tissues containing desmosomes, including stratified epithelium, which suggests that they may play specific roles. However, the roles they play are not yet fully understood. The presence of the desmocollins and the desmogleins in the desmosomes suggests that, like the cadherins, desmosomes may also play a role in cell-cell adhesion and embryogenesis. At the time of starting this project, little was known about desmosomes in development and they had only been studied in mouse preimplantation embryos, and the in developing mouse and human kidney. During mouse development, desmosomes first assemble in the early trophectoderm at the 32-64 cell stage, which is concomitant with the onset of blastocoel cavitation. The earliest desmocollin isoform to be detected at this stage is Dsc2. RT-PCR has subsequently demonstrated that DSC2 mRNA is transcribed at the 16 cell stage just prior to the onset of protein synthesis and may regulate desmosome assembly. In developing human and mouse kidney the assembly of desmosomes is associated with early tubule morphogenesis, and suggests that desmosomes may play a role during epithelial morphogenesis. The aim of this thesis was to determine the expression of desmosomal components during mouse post implantation development, in particular, the desmocollins, and contribute to the understanding of their role. The project commenced by studying the expression of the desmosomal component, desmoplakin, during post implantation development using indirect immunocytochemistry with antibodies raised against desmoplakin I and II. The presence of desmoplakin throughout embryogenesis suggests that desmosomes may play a role in epithelial morphogenesis and heart organogenesis. Expression of the desmocollin isoforms was established in post implantation mouse embryos (7 to 18 days gestation) using RT-PCR. DSC2, previously shown to be expressed from the 16 cell stage, was found to be continuously present in embryos aged from 7.5 to 18.5 days, suggesting that DSC2 is the fundamental desmocollin isoform present in desmosomes throughout development. DSC3 and DSC1 mRNA are expressed later, in embryos aged 12.5 and 15.5 days, respectively. Furthermore, the spatial expression of Dsc1 and Dsc3 was established using in situ hybridization and immunocytochemistry, and results show that Dsc3 and Dsc1 are strongly expressed during epidermal development simultaneous with specific events, including stratification and differentiation, respectively, thus suggesting they may play a morphogenetic role within the developing epidermis.
