Özet:
Organophosphates (OPs) are chemical elements that are made from mixed reactions of alcohols and phosphoric acid. The main purpose of OPs is to be used as an insecticide to wore off unwanted bugs and pest for farmers especially in developed countries such as South America and some parts of Africa and China. Unfortunately, the chemical Organophosphates can be synthesized to become nerve agents such as chlorpyrifos (CPF). CPF only become toxic in the liver when it combines with an oxygen to form CPF-Oxon. From there the CPF-Oxon inhibits an enzyme called Acetylcholinesterase (AChE). AChE job is to hydrolyze a molecule acetylcholine (ACh) to stop the buildup of the biomolecule in the body, the inhibition of AChE will reverse this metabolism process. In my research, I model the harmful effects of Organophosphates CPF/CPF-oxon in an in-vitro whole-body modeling via Physiologically based Pharmacokinetic and Pharmacodynamics (PBPK/PD). The model was built with a four brian compartment that is model after the anatomy of the brain. This research is about developing a PBPK/PD model that shows the inhibition of the enzyme acetylcholinesterase (AChE) based on the metabolism of the organonophasate chlorpyrifos (CPF).
