The purpose of this investigation was to discover whether and in what way characteristics of ion channels in rat sarcolemma change during postnatal development and to relate any such changes to the alterations in membrane permeability to sodium and potassium ions that take place in muscle membrane during development. A method of producing membrane vesicles from mammalian skeletal muscle was developed. Extensor digitorum longus muscles from neonate rats were incubated in 180 units/ml collagenase in 120 mM potassium solution at 37°C for between 30-50 minutes. This produced a clean membrane surface in the form of vesicles capable of forming high resistance electrical seals with glass patch clamp pipettes to measure single channel currents. The cell attached patch configuration was used to determine current/ voltage relationships at room temperature. Activity from several kinds of channels were recorded on videotape and data was analysed using a suite of computer programs. Single channel conductances ranged between 73-332 pS for calcium-activated potassium channels, around 160 pS for ATP-sensitive potassium channels and 58 pS for inward rectifiers. Pna/PK was found to be 0.032 for calcum-activated potassium channels. ATP- sensitive channels were observed for which channel activity decreased by 93% when 0.7 mM ATP was added to the bath solution. However, in some cases channel activity was increased upon exposure to ATP open probability rising by 40% from 55% to 95%. Age-dependent changes in the characteristics of calcium-activated potassium channels were assessed. The length of time a channel remained open once it entered the open state reached a peak between days 12-17 v4ien most developmental changes in skeletal muscle take place. In addition, open probability curves were shifted to the left with an increase in age. Overall, lower single ion channel conductances were observed in younger muscle and higher values were seen in older animals. Potassium ion permeability decreased very slightly.
