Hydnora Johannis Becc (Kausen Kasa) is a holoparasitic herb, traditionally used in modulating menopausal symptoms, treating infertility, dysentery, diarrhea, cholera and gastric ulcer in some Asian and African countries. This study investigated effects of Hydnora Johannis extract on the proliferation of H1299 cell line. Cells were plated in 96 well plates and the effects of Hydnora Johannis were evaluated using MTT assay. Treatment of H1299 with Hydnora Johannis extract for 24 hours induced a dose dependent increase in cell proliferation, increasing growth by 165%, 188%, 203%, 214% and 263% at 0.1, 0.25, 0.5, 1 and 2 mg/ml respectively, compared to the control. Paradoxically, the proliferative effects of the extract were turned into antiproliferative effects at 48-hour treatment reducing the cell growth to -74%, -65%, -37%, -17%, and 108% for 0.1, 0.25, 0.5, 1 and 2 mg/ml, respectively compared to the control. The pathways and mechanisms by which Hydnora Johannis regulate H1299 cells growth were investigated by determine the roles of Estrogen receptors (ER), Epidermal growth factors (EGFR) and Protein Kinase C (PKC) mediated signaling pathways. Hydnora Johannis Becc (Kausen Kasa) extract contains numerous constituent's alkaloids compounds which could regulate these pathways in H1299. Hence, cells were treated with Hydnora Johannis extract 0.1 -- 2 mg/ml in the presence of estrogen receptor inhibitor (Tamoxifen10microg/ml), EGFR/RTK inhibitor (Tyrphostin, 5 microM), Protein Kinase C inhibitor (Calphostin C, 5 microM) and effects on cell proliferation was determined. Tamoxifen, Tyrphostin, and Calphostin C significantly attenuated proliferative actions of 24 hours Hydnora Johannis extract treatment. The antiproliferative effects observed at 48 hours following Hydnora Johannis treatment prevented and was reverted to proliferation in the presence of these inhibitors. These results suggest that Hydnora Johannis can activate both proliferative and apoptotic pathways in H1299 cell line through activation of ER, EGFR and PKC signaling pathways. Our results call for further investigations to determine the downstream and possible transcription factors involved in this paradoxical action of Hydnora johannis extract.
