The microbiota plays various roles in host health, such as modulating non-specific host defense, including elements of the immune system, and aiding food digestion. Diet is a significant contributor to the structure of the gut microbiota. My dissertation project involves the use of an insect model to investigate the impact of diet on: 1) the ability of the microbiota to resist invasion by pathogens, and 2) the host's susceptibility to infectious diseases. Colonization resistance, or the ability of a microbial community to prevent colonization by invaders, is a key component of host defense against pathogens; as is the host's immune response. Shifts in microbial composition can result in altered colonization resistance and the activity of the immune system in response to pathogens. The objective of this study was to determine the ability of a specific group of plants and their phytochemicals to influence the insect microbiota and its immune response to pathogens. This work has clear implications for pest control and other agricultural applications, but also provides results that are generalizable and will influence how we conceptualize dietary components and their impacts on host-microbe interactions.
Larvae of Pieris rapae, the cabbage white butterfly, contain a relatively simple midgut microbiota and can be easily reared and monitored in the laboratory. The natural diet of this insect is comprised mostly of brassicas such as the cruciferous plants, Brussels sprouts, broccoli, kale and cabbage. Glucosinolates, a class of phytochemicals found within each of these plants, has anti-cancer and antimicrobial activities. Previous work has shown that sinigrin, a glucosinolate, plays a role a role in shaping microbiota structure in the P. rapae midgut and can alter colonization resistance to non-pathogenic invaders. Therefore, it is hypothesized that sinigrin impacts community invasibility by pathogens. This was tested by feeding larvae sterile artificial diet amended with sinigrin and challenging the insects with a common insect pathogen, Serratia marcescens. Additionally, I determined whether sinigrin and glucosinolate derivatives alter immune response to this pathogen.
