Özet:
The central nervous system (CNS) in mammals has no capacity for regeneration after injury. As a consequence neurons degenerate and die, partially because they are unable to maintain trophic support. In contrast, amphibians and fish have a considerable proportion of retinal ganglion cells (RGCs) that survive after axotomy and are able to regenerate. In the frog Rana pipiens, many RGCs (approximately 50% of them) die after optic nerve injury. However, the surviving cells show axonal regeneration, with eventual reconnection to appropriate brain targets. For this reason the frog visual system is a good model to understand the role of neurotrophins in survival, regeneration and synapse formation during reconnection. Our focus is to try to understand the regeneration process; and to study how neurotrophins affect the ultrastruture of the regenerating optic nerve. The goal of this project is to determine the effects that neurotrophic factors, such as ciliary neurotrophic factor (CNTF), fibroblast growth factor (FGF-2), and brain-derived neurotrophic factor (BDNF) have on the speed, number and distribution of the regenerating axons after optic nerve crush. In addition, we propose to study the ultrastructural features of the regenerating optic nerve and the differences that treatment with CNTF and FGF-2 has on the cells populations in the nerve. We plan to pursue the following specific aims: Aim 1. To determine the effects of CNTF, FGF-2 and BDNF on axonal regeneration. Hypothesis: Application of neurotrophic factors (CNTF, FGF-2, BDNF) increases the speed of axonal regeneration and increases the number of regenerating axons in the optic nerve. Aim 2: To determine how the application of CNTF and FGF-2 affect the macrophage cell populations in the regenerating optic nerve. Hypothesis: Application of CNTF and FGF-2 increases the total number of macrophages in the lesion and the regenerating zones. This study will contribute to our understanding of the factors and signals that influence regeneration after optic nerve injury in the amphibian Rana pipiens. These mechanisms are likely to be the same in all animals, so this study may indicate potential therapeutic applications of these neurotrophic factors.
