Since their discovery in 1953 (Rowe et al., 1953), and. particularly over the last decade, the adenoviruses have been extensively studied using diverse biological, biochemical, and genetical approaches (for recent reviews see Tooze, 1973; Philipson and Lindberg, 1975; Ginsberg and Young, 1976; Williams, 1976). Over 80 serotypes of adenoviruses, of human, simian bovine and avian origin have been described (Wadell, 1970) and the 31 serotypes of human origin may be subdivided into three groups on the basis of serological tests (Rosen, 1960) In their natural hosts adenoviruses cause respiratory diseases and may also be involved in disorders such as enteritis and conjunctivitis. Human adenovirus type 12 was shown in 1962 to cause tumours when injected into newborn hamsters (Trentin et al., 1962), and a number of other adenoviruses have since been shown to be oncogenic in newborn rodents, or to transform rodent cells in vitro. There is no evidence, however, that the adenoviruses can cause tumours in their natural hosts. The wide interest which has been shown in the adenoviruses may be attributed to a number of their general properties: 1. A variety of serotypes, with a spectrum of biological activities, are available for study. 2. Adenoviruses grow to high titre in tissue culture and are easily purified. 3. Large excesses of the major capsid proteins are pro duced in infected cells, and these may be studied both biochemically and immunologically. 4. Recently developed accurate plaque assay systems have facilitated the isolation of adenovirus mutants, and their use in genetical studies. 5. Adenoviruses are the only human agents which, on the basis of tumour production in animals, have so far been shown to have onogenic potential.
