The control mechanisms of lymphocyte traffic in the mouse, were investigated. Lymph node cells, a representative sample of the recirculating pool of lymphocytes, were labelled with radioactive isotopes. (51Cr or [3H]-Adenosine) and their migratory patterns studied after i.v. transfer into syngeneic recipients. Information about the nature and role of different components of the lymphocyte membrane in the process of continuous recirculation between blood and lymph, may be gained by studies with substances which modify the structure of the membrane. Therefore, the effects on cell traffic of agents such as, trypsin, neuraminidase, plant lectins (Concanavalin-A and Phytohaemagglutinin), lipopolysaccharide, dextrans derivates and phospholipases, were investigated, All these agents were shown to alter significantly the localization of labelled lymphocytes. The results obtained fail to demonstrate the existence of lymphocyte receptor specific for the high endothelial cells of the PCV of the lymph node and fail to support the exclusive role of the interaction lymphocyte-PCV as the only mechanism regulating the nature and rate of lymphocyte recirculation. An alternative mechanism for the control of lymphocyte traffic is proposed. According to the advanced hypothesis, lymphocyte traffic, its speed and characteristics, are determined by continuous cell to cell interactions occurring between the circulating lymphocytes and resident cells in lymphoid and non-lymphoid organs. The nature of such interactions is probably conditioned by general properties of the lymphocyte, such as, adhesion, electric charge, membrane fluidity, etc., rather than by the presence of multiple specific receptors on the cell surface. The relevance of the present findings to the understanding of the biological significance of lymphocyte recirculation is discussed.
