The first section of this thesis considered the effects of calcium on lead absorption from the gastrointestinal tract and the elucidation of the mechanism of uptake of lead from the gut. Al. The effects of calcium on lead absorption as lead chloride and 203 Pb chloride was studied in rats. Doubling of dietary calcium with calcium orthophosphate significantly depressed lead absorption as assessed by 203Pb retention, tissue lead concentration, excretion of urinary delta aminolaevulinic acid (ALA) ajid activities of the enzymes ALA dehydratase (ALA.D) and ferrochelatase (FERRO.C). Similar effects on lead retention were shown by the calcium salts; chloride; carbonate; orthophosphate; sulphate; gluconate and glycerophosphate. Calcium was found to be most effective in depressing lead retention when administered immediately prior to lead, whilst the fact that increased systemic calcium had no effect on lead retention indicated that the effect of calcium in reducing lead retention was mediated through the gastrointestinal tract. The second section of this study has considered haem biosynthesis and the effects of lead upon it.
