Triple-negative breast cancer (TNBC) is aggressive cancer found in women that mostly have poor clinical outcomes. African-American women have a lower incident rate of this type of breast cancer compared to women of European descent. However, African American women have a higher mortality rate despite their overall breast cancer incidence for this specific type of breast cancer being lower. This discrepancy poses a major health concern and compels researchers to explore more effective treatment options than those currently available. Research has shown that chemoprevention of breast cancer with the use of natural compounds have shown promise because of low toxicity and fewer side effects compared to synthetic compounds. Apigenin (4'5, 7 trihydroxyflavone) is a flavonoid commonly found in many fruits and vegetables such as parsley, chamomile, celery, and kumquats. Apigenin is derived from Apium genus in Apiaceae (celery, carrot, or parsley family, also known as Umbelliferae). In the past few decades, apigenin has been recognized as a cancer chemopreventive agent. Studies have shown that apigenin contains anti-carcinogenic properties through apoptosis and induction of autophagy in several human cancer cell lines. Apigenin has demonstrated to be the most effective phytoestrogen that strongly inhibits the growth of breast cancer cells. Breast tumor cells differentially express tumor necrosis factor (TNF) and an abundance of cytokines. Research has shown that TNFalpha that is present in breast tumor cells support tumor growth. Of the cytokines, GRO and GROalpha are known to be potential markers for cancer, and they are associated with tumor progression for breast cancer. Preliminary studies conducted in our lab have proven that apigenin can suppress TNFalpha induced GRO and GROalpha from triple negative breast cancer cells Based on these facts, we hypothesize that apigenin may regulate GRO and GROalpha cytokine release in TNBC cells. The overall rationale for this proposed research is, if proven successful, may provide therapeutic targets to inhibit metastasis and prolong patient survival.
