Ganoderma applanatum antigens can induce human sensitization and has been described as allergenic. The allergenicity of Ganoderma spp. has been much more studied than any other basidiomycete. Allergens from fungi have been linked to respiratory and skin symptoms and can trigger asthma exacerbations or worsen symptoms in sensitized individuals. Fungal allergy has been shown to play an important role in severe asthma.
\A total of 63 human sera were tested and divided in two groups according to their serological reactivity. The group 1 included those serums reactive to one or more allergens, including fungi and the second group included those sera without reactivity to the allergens in the panel. Although the ELISA immunoreactivity in group-1 was 36% higher than group two, meaning that the allergen reactivity of group-1 was 36% greater than in group-2, there was no statistical significance as determined with the Wilcoxon rank sum test. Concluding that the ELISA results presented a non-normal distribution, that may be due to the small n of group 2 (13 samples) compared with group 1 (51 samples). Also, we attribute to the complexity of the Cytoplasmic Mycelial Crude Extract (CMCE) sample, the reason that did not allow serum IgE to discriminate the allergenic proteins of Ganoderma applanatum mycelium in assays like ELISA or Western Blots. As the CMCE is a heterogeneous sample, this makes it to be a non-optimal sample to work with or to be used for sero-diagnostic purposes, instead, the proteins must be fractionated or isolated and purified in order to work with.
Next, we studied the proteome of the crude cytoplasmic extract of the mycelium and the spores using electrospray ionization (ESI) technique in tandem mass spectrometry and overlapped both proteomes to identify shared allergenic protein(s) of the mycelium and spores of Ganoderma applanatum (G. applanatum). As a result of this analysis three proteins were identified: polyubiquitin, catalase and calmodulin which are common to both, mycelium and spores of G. applanatum. The finding of catalase is consistent with the findings of Vila-Hereter et al. 2017, who recently identified this protein (named as a Hypothetical protein) by western blot and MS/MS in the spores of G. applanatum and demonstrating that this molecule could constitute an important marker of fungi-mediated allergy. Our data also suggest that the Protein 2 named as a Hypothetical protein by Vila-Hereter et al and the catalase identified in our investigation, are the same molecule, inferring that the catalase of the G. applanatum mycelium may have allergenic characteristics as well. We conclude that catalase is an important protein with regards to allergenicity of G. applanatum , representing an opportunity to be used as biomarker in diagnostic panels or to develop specific target therapies or treatments.
