Wild-type Sendai virus is pneumotropic while its variant, F1-R, is pantropic. Two phenotypic differences in F1-R are attributed for its ability to cause a systemic infection: 1) Enhanced cleavability - mutations in the fusion (F) gene. The F gene, encoding a fusion glycoprotein, functions in fusing the viral envelope with the host membrane to mediate viral entry, and requires proteolytic cleavage by a lung specific protease. In contrast, F1-R F is cleaved by ubiquitous proteases allowing the virus to replicate in multiple tissues. 2) Bipolar Budding --mutations in the matrix (M) genes. Wild-type virus buds from the apical domain of the cells. F1-R, however, has two mutations in the M gene resulting in an expressed matrix protein that disrupts the microtubule network and polarized transport leading to budding from both apical and basolateral domains. Thus wt virus is released into the lumen of the respiratory tract, and F1-R is released both into the lumen and the basement membrane where it may access the bloodstream for dissemination. Together with enhanced cleavability and bipolar budding, it can reach various tissues and replicate as well. To identify the specific mutations responsible for the systemic phenotype of F1-R, Reverse Genetics was used to generate variant RGV-0 virus that contains all F1-R F and M gene mutations and can cause a systemic infection. For this study, RGV-19, was constructed which contains F1-R F and only one M gene mutation that would test the contribution of the Matrix protein to the systemic phenotype of F1-R.
