Injury, infection and underlying genetic factors can contribute to various neurodegenerative diseases. Neurodegenerative disease can impact cognition, motor ability and also contribute to premature death. In the case of Alzheimer's disease, neurodegenerative activity is associated with cognitive and motor impairment due to accumulation of the beta-amyloid (Abeta) peptide around neurons within the brain. The beta- amyloid protein is produced due to the improper proteolysis of cell surface protein known as amyloid precursor protein (APP). Identification of cellular mechanisms and genes involved in the regulation of APP synthesis in neurons will further our understanding of the role this protein plays in neuronal cell function and neurodegenerative diseases.
In this study, laminin, a basement membrane glycoprotein that is a powerful juxtacrine stimulator of neurite outgrowth, was utilized to differentiate cultures of pheochromocytoma cells (PC12) and investigate APP expression within this neuronal cell line. PC12 cells were initially primed with the Nerve Growth Factor (NGF) and grown on laminin to determine its potentiating effects in neuronal differentiation. Changes in the cells' morphology were observed prior to cellular collection and RNA isolation. Levels of APP gene expression were determined by reverse transcriptase PCR (RT-PCR) in which a cDNA library was created and subjected to PCR amplification. The PCR product was analyzed by gel electrophoresis to determine the transcriptional effects of laminin on APP specifically. The RT-PCR product was compared to positive and negative controls such as beta-actin and tyrosine hydroxylase at two, four, six and eighteen hours. Cells that were grown on laminin exhibited a steady decline after two hours in APP expression.
